Establish ranged superiority and throw stuff at it until it crumbles into dust, then burn the dust until nothing remains but a puddle of molten rock. It takes a long time, requires a massive concentration of valuable assets, but it's the only way that works.

I don't think Anthony becoming a queen would be in the Colony's best interest. In order to become a decent queen he would have to invest significant skill points and biomass into his egg laying organ. This would negatively impact his other aspects that he is rather fond of E.G. his shiny carapace, magic, gravity gland, business district... All that compromise would likely make him ineligible to become an ancient similar to the Mother Tree who lost the interest of the dungeon when she created her family. Of course if it's just a last resort, then investing all his resources into not letting the Colony die and keeping himself alive would be in the Colony's best interest. He can consider becoming a queen after the Colony is wiped out.
