Bro Tsunade and Minato both know about Kushina's Jinchuriki status, and Kyuubi was required to be sealed in a Uzumaki because they have special chakra and life force to contain the Kyuubi plus reaper death seal have no condition on this and Kushina had Kyuubi sealed in her as a teenager, with Mito having Kyuubi sealed in her as an aged Adult (Kyuubi was sealed in Mito after Hashirama vs Madara, after which few years Hashirama died and him spoiling Tsunade with gambling addiction means that Mito was already a gradnma).
