Well preferably the core because one weak spot is better than two but then again if that happens, all the enemy has to do is take out that one spot and both the consciousness and the core dies.
If the core Spark suffered fatal damage or was removed, the tiny Sparks within each cell would be extinguished, resulting in the death of the Transformer. Similarly, if the head was destroyed, eliminating consciousness, the Transformer would also die.
